PikoReal Software Release Notes Version 2.1 -> 2.2 major changes Relative Quantification 

Corrected calculations to use geometric mean when averaging relative quantities

Error bars added to relative quantification bars

Control error bar size (1 – 3 times standard deviation)

RQ bars sorting option added

Hide reference genes from RQ graph

Min RQ and max RQ added to results table (error bar upper and lower values)

High Resolution Melting 

Automatic clustering algorithm added

Matlab routines installed for the automatic clustering algorithm

Colors assigned to each cluster automatically

Control settings for the automatic clustering algorithm

Distance to cluster average added to the results table

Ability to force calls to clusters

Ability to rename clusters

Default call mode is difference graph instead of scatter plot

Difference graph is visible without defined standards

Default temperature range is applied on first analysis

Scatter plot is detachable

Melt Curve 

Positive derivative calculation added

On/off control for the slope correction added

Cq determination 

Automatic threshold button changed to tick box

Automatic threshold calculated automatically with standalone runs imported from USB

Color calibration values applied immediately post-run when using imported plate layout

Plate layout 

Sample Type colors improved

Ctrl+z keyboard command activates undo function

Protocol 

Root protocol properties of completed runs are locked

Time and temperature decrements are handled in the same manner

General 

Replicate column added to results spreadsheets (Cq and Melt)

Well selector tooltip available at all times

Default name when saving a new file is current date and time

Default folder in Settings set to C:\Thermo\PikoReal2\userData

Shortcut to examples folder added to userData folder

Improved layout in example files

Additional example files for HRM and protein melt added

Updated protocol files

Manuals added to installation location C:\Thermo\PikoReal2\manuals

Desktop shortcut to instrument and software manual added during installation

Graph colors defined by well match RGB color and are not dependent on fluorophore selection

Protocol log import from USB standalone run supported (requires firmware support)

Known issues remaining 

Exported absolute quantification standard curve equation (Log quantity on Y axis and Cq on X axis) is different from the default analysis equation (Log quantity on X axis and Cq on Y axis)

Use flip X and Y axis to obtain same equation in the software and the export

Detached graph may be obscured by main window

No notification if user tries to save on read only file

Replicate borders do not disappear when using undo function in plate layout

Version 2.0 -> 2.1 major changes General 

User interphase color scheme changed

Language support for Japanese and Chinese

Graphs included in the Excel export

High Resolution Melting 

Completely new analysis module added to the software

Difference plot analysis

Scatter plot analysis

Protocol 

Default lid temperature setting changed from automatic to manual 105 degrees Celsius (in the firmware version 2.2.20 automatic settings for lid temperature are too low)

Cq determination 

Default baseline method changed from Curve Minimum to Trend

Version 1.5 -> 2.0 major changes General 

Complete modernization of the user interface

Virtual Pipetting Tool

User manual added

Improved analysis modules functionalities (absolute quantification, melt curve analysis, allelic discrimination, relative quantification)

Example files added

Color calibration utility added
